خلاصه مقاله: Cystic echinococcosis (CE) is caused by the larval stage of Echinococcus granulosus. The disease is cosmopolitan and Iran is a highly endemic area for CE. This parasite exhibits high genetic diversity which can be related to the life cycle, transmission, pathogenesis and effect on treatment and control. The aim of this study was to determine phylogenetic relationship and intra-genotyping variation of E. granulosus spp in a vast area in southwest of Iran (SWI). 72 hydatid cyst isolates of intermediate hosts were collected. The sequence analysis of cox1 gene showed that three genotypes G1, G3, and G6/G7 were found. G1 was the most common genotype in SWI. Twenty-six haplotypes were identified for cox1 genes. Also, 254 nucleotide translocations were observed among cox1 haplotypes, including 140 (55%) transition and 114 (45%) transversion. According to the phylogenetic tree topology, although cox1 gene similarities can be shown for some isolates of G1 in some vast areas but in overall G1 strain showed a heterogeneous population worldwide. High similarity was shown between G3 isolates from different countries and hosts. The only SWI G6/G7 haplotype were taken a distant position in E. canadensis clade that shows the notable difference of this haplotype from other isolates from Iran and other countries. The presence of G6/G7 strain in SWI may be due to transmission of strain from other regions or role of camels/wild boar or other possible hosts in the expansion of this genotype in SWI. The results of the present study can be useful tips to CE control programs, molecular epidemiology, and phylogenetic studies in Iran and other countries for future goals.
