Background and Aim

Inflammation and stress oxidative are two major factors that lead to the spreading secondary phase of spinal cord injury(SCI), so reducing these factors is an effective way to control the SCI. This study intended to evaluate the protective effect of alcoholic extract of Moringa peregrine leaf on the spinal cord against inflammation and oxidative stress damage induced by SCI.

Methods

thirty-five adult male Wistar rats were accidentally separated into five groups: control, control+ M. peregrine(150 mg/kg for fourteen days), sham(laminectomy)+saline, SCI, and SCI+ M. peregrine(150 mg/kg for fourteen days). After the injury, until fourteen days all animals were assessed by BBB (Basso, Beattie, and Bresnahan) for hind limb motor function. For evaluation of antioxidant enzymes(Superoxide dismutase (SOD), catalase (CAT), and Glutathione peroxidase(GPx) ), oxidative stress markers malondialdehyde (MDA), and Total antioxidant), inflammatory factors (interleukin-1β(IL-1β), interleukin-6(IL-6), interleukin-18( IL-18) and tumor necrosis factor-α(TNF-α), and nuclear factor kappa B (NF-κB) in spinal cord tissue all animal were sacrificed on the fourteenth day.

Results

Intraperitoneal injection of M. peregrine alcoholic extracts considerably decreased the expression of pro-inflammatory cytokines such as TNF-a, IL_1b, IL_6, IL_18, attenuated level of MDA, raised levels of SOD, GPx, CAT, and total antioxidant in spinal cord tissue; Furthermore, the level of NF-kB in the SCI+ M.peregrine group was significantly lower than SCI group. therefore motor function was improved in the treatment group compared to the SCI group.

Conclusion

This conducted study shows that alcoholic extract of M. peregrine leaf has a neuroprotective effect against inflammation and oxidative stress induced by SCI. as a result, motor recovery in the treatment group is better than the untreated group.
