Evaluation of antifungal effects of hydro-alcoholic extract of Nigella sativa and Urtica dioica on fungal agents isolated from clinical specimens

Background and Aim
Due to the indiscriminate use of chemical drugs to treat infectious diseases, multidrug resistance of human pathogenic microorganisms has developed and caused many problems that always threaten human health. The occurrence of fungal species resistant to antifungal drugs and also their numerous side-effects have persuaded researchers to work on new therapeutic methods For this reason, today, medicinal plants have received more attention than ever in the treatment and pharmacological process; So far, many researchers have tried to find natural antifungal agents with fewer side effects In this regard, the antifungal activity of some medicinal plants has been proven Therefore, this study was designed to evaluation of antifungal effects of hydro-alcoholic extract of Nigella sativa and Urtica dioica on fungal agents isolated from clinical specimens.
Methods
In the current study, antifungal effects of hydro-alcoholic extract of Nigella sativa and Urtica dioica against 11 Candida isolates (Candida albicans, Candida parapsilosis), 13 Aspergillus isolates (Aspergillus flavus, Aspergillus terreus) and 5 Mucoral agents (Mucor circinelloides, Rhizopus oryzae) were examined using broth microdilution method. The tested fungal isolated were associated with different concentrations of the plant extracts, and then, the Minimum Inhibitory Concentration (MIC) of these extracts were determined separately along with amphotericin B.
Results
Both soaked and sox let hydro-alcoholic extracts of Nigella sativa and Urtica dioica showed antifungal effects against Candida, Aspergillus and Mucorales' isolates. The MIC90 values of soaked and sox let hydro-alcoholic extracts of Nigella sativa in C. albicans and C. parapsilosis were 25, 12.5mg/ml respectively. For Aspergillus flavus in both methods were 25mg/ml but, in Aspergillus terreus in soaking and sox let methods were 12.5 and 25mg/ml respectively and in mucoral species (Mucor circinelloides and Rizopus. oryzae) were obtained 12.5mg/ml. The MIC90 values of soaked and sox let hydro-alcoholic extracts of Urtica dioica in Candida and mucoral species in both methodes were 12.5mg/ml, But in the Aspergillus species were obtained 25mg/ml.
Conclusion
The Hydroalcoholic extracts of Nigella sativa and Urtica dioica have acceptable antifungal activity on Candida, Aspergillus, Mucor and Rhizopus species. Urtica dioica extract has better efficacy against Candida species and Mucorales.
