Background and Aim

Mycoplasma species threaten reproduction and fertility worldwide. The lack of a sensitive and accurate diagnostic method has led to the neglect of the role of these bacteria in infertility. The aim of this study was to identify and compare the frequency of Mycoplasma isolates in fertile and infertile women using Real-Time PCR and melting curve analysis.

Methods

In this case-control analytical study, cervicovaginal swap samples were taken from 140 patients referred to two teaching hospitals in Isfahan. Using a new primer designed to amplify 16S rRNA gene and real-time PCR technique, several types of Mycoplasma and Ureaplasma in two fertile (n = 70) and infertile (n = 70) groups were examined.

Results

The frequency of Mycoplasma isolates in infertile and fertile groups was as follows respectively: Mycoplasma hominis 17.4% (n = 12), 7.1% (n = 5), Mycoplasma genitalium: 2.8% (n = 2), 0 % (n=0) ureaplasma parvum 17.4% (n = 12), 5.7% (n=4), Ureaplasma urealiticum 10% (n = 7), 4.2% (n = 3), coinfection 22.8% (n = 16%) and 2.8%( n =2). The designed primer made it possible to simultaneously identify Mycoplasma species in cervicovaginal specimens.

Conclusion

In the present study, the frequency of Mycoplasma and Ureaplasma species in the infertile group was more than 2 times that of the fertile group. Therefore, it is important to pay attention to the role of these infectious agents in infertility.
