Background and Aim

Background: Liver fibrosis is the result of chronic liver injury and a major cause of morbidity and mortality in the world. The present study was done to investigate the protective effect of resveratrol on liver fibrosis, focusing on the possible involvement of TGF-β signaling through Smad2. Hepatic stellate cells (HSCs) activate during chronic liver injury, express more transforming growth factor-beta (TGF-β), Collagen (COLA1), and actin-alpha smooth muscle (αSMA) that lead to hepatic fibrosis. Aim: This study investigated the antifibrotic effects of resveratrol on palmitic acid-activated human HSC and its mechanism in vitro.

Methods

Methods: First, the human HSCs (LX-2)were treated with palmitic acid (125μM) for 24 hours and then with resveratrol for 24 hours. Total RNAs were extracted, reversely transcribed into cDNA, Quantitative Real-time PCR and western blot were performed.

Results

Results: The mRNA expression levels of TGF-β, COL1A, αSMA genes, and the level of smad2 phosphorylation in palmitic acid-activated LX-2 treated with resveratrol were significantly reduced compared to palmitic acid-activated LX-2.

Conclusion

Conclusion: Resveratrol is effective in reducing the expression of fibrogenic genes in the activated LX-2 treated with palmitic acid through downregulation of the TGF-β/smad2 signaling pathway. Therefore, resveratrol possesses significant antifibrotic properties in hepatic fibrosis
